Dunbar United, Musselburgh Athletic and Tranent are all involved in the first round of the competition.
Bo'ness Athletic are the visitors to New Countess Park while Darvel head to the Olivebank Arena.
Tranent face an intriguing trip to Dundee North End.
Meanwhile, Preston Athletic and Ormiston Primrose are looking for three points.
Preston head to Kinnoull in the East of Scotland Football League (EoSFL) First Division while Ormiston welcome Edinburgh United in the Third Division.

A PLACE in the second round of the Scottish Cup is up for grabs for Musselburgh Athletic and Darvel.
Liam Burns and his side gave Clyde a fright in the competition last year before going out in extra time.
Meanwhile, Darvel made headlines when they knocked Aberdeen out of the tournament a couple of years ago.
Musselburgh play their football in the top tier of the EoSFL while Darvel are in the Premier Division of the West of Scotland Football League (WoSFL).

HEADLINES were made last year when Dunbar United knocked East Fife out of the Scottish Cup.
Now, they will be looking to make it to the second round again when they welcome Bo'ness Athletic.
The visitors have enjoyed back-to-back promotions to reach the First Division of the EoSFL.
However, they lost on their last visit to East Lothian when Preston Athletic came out on top at the weekend.

JONNY Stewart will get his first experience of the Scottish Cup as a manager this afternoon as Tranent travel to Dundee North End.
The Midlands League champions come into the contest boosted by a 25-0 win over Whitehills.
However, it will be a step up in class when Scottish Lowland Football League side Tranent visit.
The Belters will be favourites but must play well to prevent them being on the wrong end of a cup shock.
Kinnoull v Preston Athletic
East of Scotland Football League First Division

VICTORY over Bo'ness Athletic last weekend continued Preston Athletic's unbeaten start under Stevie McLeish.
Now, they are looking for another three points.
Kinnoull sit second bottom of the table with just one win from their opening eight league games.
Three points for Preston could lift them above a number of clubs and into mid-table.
Ormiston Primrose v Edinburgh United
East of Scotland Football League Third Division
Kick-off 2.30pm
GOAL difference is all that separates Ormiston Primrose and Edinburgh United in the middle of the Third Division.
Three wins and three defeats from the opening seven games has both teams locked together on 10 points.
A win for either side could be enough to put them into the promotion places.
A tight contest could be on the cards.
